SUMMARY:

Cyanobacteria have in their composition several pigments (e. g. carotenoids, phycobiliproteins, and photoprotective compounds) that are potential substitutes for synthetic pigments used in the food, cosmetic, textile and pharmaceutical industries. These microorganisms can be found in the most diverse environments and the Atlantic Forest of southern Bahia presents conditions to support biodiversity with the potential for the production of pigments not yet characterized. Thus, the present research aims to select cyanobacteria isolated in reservoirs located in the Atlantic Forest of southern Bahia, with potential for pigment production. To this end, after registering access to the national genetic patrimony in SISGEN, the samples collected in reservoirs were subjected to isolation using the techniques of “fishing” and spread plate in BG-11 and ASM-1 medium. Incubation took place in BOD with a photoperiod of 12 h light and 12 h dark, light intensity of 20 µmol·m-2·s-1 , at 25 ºC. The microorganisms were identified at the genus level based on morphology, and cultured for 14 days in 300 mL of the respective isolation media and conditions, plus UVA/UVB light. Growth was monitored by spectrophotometric quantification of chlorophyll (via ethanol extraction) every 48 h. At the end of the cultivation, the pigments were extracted with solvents (ethyl acetate for nonpolar and saline phosphate buffer for phycobiliproteins) and detected by spectrophotometry. The selection was based on pigment concentration and biomass accumulation. The selected isolates were cultivated in a bubble column photobioreactor, with previously characterized hydrodynamics, and the kinetic parameters were determined: maximum biomass concentration and biomass and pigment productivity. In addition, the technological prospection of the production of pigments by cyanobacteria was carried out through patent research in national and international repositories. Altogether, 17 cyanobacteria capable of growing in ASM-1 or BG-11 media. The six genera found are from the orders Synechococcales (02) and Oscillatoriales (15). Two isolates were selected: Phormidesmis sp.1 Turicchia et al 2009 and Leptolyngbya sp.1 Anagnostidis et Komárek 1988. Phormidesmis sp.1 showed a dry biomass accumulation of 1.0 ± 0.15 g/L, 54.39 ± 9.40 mg/g phycocyanin, and 60.07 ± 11.33 mg/g allophycocyanin. The percentage of phycocyanin in the biomass is similar to that of Arthrospira platensis and the allophycocyanin content is above that found in the literature (44.08 mg/g). In Leptolyngbya sp.1, a maximum concentration of phycoerythrin of 55.78 ± 3.92 mg/g was detected, in addition to allophycocyanin (46.70 ± 11.07 mg/g) – levels similar to those of optimized extractions in species used in the studies of phycoerythrin production from cyanobacteria. These isolates presented concentrations significantly different from the others, in order to be used in the following stages of the project. In photobioreactor, Phormidesmis sp.1 showed maximum productivity of biomass, phycocyanin, and allophycocyanin of 0.127 gdry mass/L/day, 6.4, and 7.75 mg/L/day, respectively. The total biomass accumulation was 1.88 g/L. In the patent search, 556 patents were retrieved, of which 119 were selected. The countries that have the most deposits are Japan (40), China (36), and the USA (20), with 32% of deposits made in the last 5 years. The main holder is the company Dainippon Ink & Chemicals, which demonstrates, a real interest in the use of these pigments in the cosmetic and textile sector
