SUMMARY:

Tegumentary leishmaniasis (TL) is a neglected zoonotic infectious disease that persists as a public health problem, with thousands of cases reported annually, caused by intracellular protozoa of the genus Leishmania. Current treatment is based on old drugs that have significant side effects and are difficult to administer. The possible discovery of new molecules could lead to the formulation of a new therapeutic strategy for topical use and low toxicity for patients with TL. However, it is important to remember that prospecting for biomolecules is a costly and time-consuming process, always considering the maintenance and conservation of biodiversity. This study aims to evaluate the potential of extracts and their sub-fractions from Piper macedoi to generate phytopharmaceuticals for the treatment of TL (Leishmania amazonensis). P. macedoi will be collected from a fragment of Atlantic Forest in the municipality of Teixeira de Freitas. The dried plant will be subjected to exhaustive extractions in an ultrasonic bath with the solvents hexane, dichloromethane, ethyl acetate and methanol. The extracts will be fractionated using a chromatographic column and then characterized using gas chromatography coupled to a mass spectrometer. L. amazonensis promastigotes with the LacZ gene encoding the β-gal enzyme will be grown in Schneider medium at 26ºC, while canine macrophages (DH82) will be grown in RPMI 1640 medium at 37ºC and 5% CO2. The parasites will be opsonized with canine serum and then infected with DH82 at a ratio of 10:1 parasites/cells. For the test, the extracts will be diluted in RPMI at concentrations of 0.01, 0.1, 1, 10, 100 and 1000μg/mL. After the reaction, Chlorophenol red-β-D-galactopyranoside (CPRG) will be added, the still living parasites will cleave the CPRG into the phenol red chromophore, which can be detected by spectrophotometry at 570nm. The cytotoxicity test will then be carried out using MTT, which is reduced to formazan by metabolically viable cells. The tests will evaluate the percentage of inhibition, CI50 and CC50. Thus, we hope to publish the scientific findings and find a new drug candidate for the treatment of tegumentary leishmaniasis.
